Salvianolic acid A exhibits anti-tumor activity in nasopharyngeal carcinoma cells through regulation of Pin1

چکیده
Background: Nasopharyngeal carcinoma (NPC) remains as a devastating malignancy due to a lack of effective therapy for recurrence and metastasis. Salvianolic acid A (SAA), a compound extracted from Salvia miltiorrhiza, possesses a variety of biological properties such as antioxidant, antithrombotic, myocardial ischemia protection and anti-hepatic-fibrosis. However, the effect of SAA on NPC cells has not been clarified yet. Methods: NPC cell lines and immortalized nasopharyngeal epithelial cells NP69 were treated with SAA at different concentrations (0-40 μM). Series of relevant experiments were carried out to test the effect of SAA on the proliferation and migration ability of NPC and NP69 cells Pull-down assay and Western blot were used to find out the possible molecular target of SAA. Results: Our results indicated that SAA could inhibit the proliferation of NPC cells in a dose and time dependent manner. And SAA was able to weaken the clone-forming ability as well. Meanwhile, the migratory and invasive capabilities were also attenuated. Moreover, SAA didn’t exhibit any cytotoxicity on immortalized nasopharyngeal epithelial NP69 at the same treatment condition. Furthermore, high concentration of SAA could promote the apoptotic of CNE2 and HNE1 cells dramatically. The results of in vitro pull down assay showed that SAA might bind with Pin1 directly. And the results of western blot indicated that SAA could down-regulate the expression level of cyclin D1, p53, p21 and Bcl-2, which are downstream substrates of Pin1. Conclusions: SAA exhibits anti-tumor activity in NPC through down-regulating Pin1 and it may be a potential drug for NPC.

متن کاملOverexpression of PIN1 Enhances Cancer Growth and Aggressiveness with Cyclin D1 Induction in EBV-Associated Nasopharyngeal Carcinoma
BACKGROUND Nasopharyngeal carcinoma (NPC) is a peculiar Epstein Barr virus (EBV)-associated malignancy that is prevalent in South-East Asia. Peptidyl-prolyl cis-trans isomerase NIMA-interacting 1 (PIN1) isomerizes specific phosphorylated amino acid residues, which makes it an important regulator in cell survival and apoptosis. In this study, we investigated the contribution made by PIN1 in NPC ...

Background: Overexpression of Rad51 protein in many tumor cells has been proven to increase radioresistance and can be related to the resistance of chemosensitivity of tumor cells. This preliminary study was conducted to determine the relationship between the Rad51 expression level in nasopharyngeal carcinoma and the response of the treatment based on the measurement o...
متن کاملInhibition of the prolyl isomerase Pin1 enhances the ability of sorafenib to induce cell death and inhibit tumor growth in hepatocellular carcinoma
Hepatocellular carcinoma (HCC) is the sixth most common cancer, but is the second leading cause of cancer deaths, partially due to its heterogeneity and drug resistance. Sorafenib is the only medical treatment with a proven efficacy against advanced HCC, but its overall clinical efficacy is still modest. Therefore, a major challenge is how to improve its therapeutic efficacy. The unique prolyl ...
متن کاملNiflumic acid exhibits anti-tumor activity in nasopharyngeal carcinoma cells through affecting the expression of ERK1/2 and the activity of MMP2 and MMP9.
Niflumic acid (NFA) was known to inhibit cell proliferation or migration in several types of cancer. However, the function of NFA in human nasopharyngeal carcinoma (NPC) cells was not clarified. The proliferation of NPC cell line CNE-2Z cells with NFA treatment was detected using the cell counting kit-8 method and transwell assay was employed to assess the effect of NFA on the CNE-2Z cell migra...
